Aligning TDC flanges before bolting

Can I use drift pins/C-clamps to hold this TDC alignment after I re-support the run?

I'm at this TDC/Ductmate joint. The gasket is on one flange face and the duct is close, but the holes/flange need help holding alignment. Do you want me to re-support and level it first, then use drift pins at the corners and C-clamps on the flange while I start the corners/cleats?
